  • Molekulare Bildgebung
  Im Rahmen dieses Projektes sollen verschiedene "Zelltracking"-Experimente durchgeführt werden. Dabei soll ein "online-Monitoring" markierter Zellen durchgeführt werden. Hierfür kommen zwei verschiedene Verfahren in Frage: Zum einen können Zellen mit Perfluorcarbonen (PFCs) beladen werden, um sie dann mittels kombinierter 1H/19F-MRT im Organismus als "Hot Spot" nachzuweisen. Zum anderen können die Zellen mit superparamagnetischen Eisenoxid-Partikeln (SPIO, superparamagnetic iron oxide) beschickt werden. Diese fluktuieren aufgrund ihrer geringen Größe (ca. 30 nm) im Magnetfeld und erhöhen durch die damit verbundene, sich ständig ändernde Magnetisierungsrichtung die lokale Inhomogenität des Magnetfelds. Die Folge ist ein örtlich verstärkter T2*-Effekt, der sich im MR-Bild als hypointense Region zeigt, so daß indirekt auf die Anwesenheit der markierten Zellen geschlossen werden kann.
  • Angiographie
  Ziel dieses Projektes ist es, die Bildqualität für angiographische Untersuchungen weiter zu verbessern und Veränderungen in der Gefäßstruktur volumetrisch zu quantifizieren. Die optimierten Protokolle sollen dann in einer Versuchsreihe zur Gefäßneubildung in CD73-/--Mäusen eingesetzt werden, um die Rolle von extrazellulär gebildetem Adenosin bei der Arterio- bzw. Angiogenese zu verifizieren.
  • Plaquedarstellung
  Während in voranstehendem Projekt luminale Veränderungen im Gefäßsystem beobachtet werden sollen, steht hier die Untersuchung der Gefäßwand im Vordergrund. Hierfür soll mittels Black-Blood-Sequenzen ein Kontrast zwischen Wand und dem Inneren des Gefäßes erzeugt werden, so daß sich Veränderungen in der Architektur der Gefäßwand über die Zeit verfolgen lassen. Darüber hinaus soll untersucht werden, ob sich durch parallele Gabe von PFCs die entzündlichen Prozesse im Plaque mittels kombinierter 1H/19F-MRT nachweisen lassen. Nach der Etablierung dieser Methoden soll dann die Arteriosklerose-Entwicklung in CD73-/--Mäusen bzw. CD73-/-/ApoE-/--Doppelmutanten verglichen werden.
  • Spektroskopische 19F-Bildgebung
  Alle bisherigen Versuche zur Bildgebung inflammatorischer Prozesse mittels 1H/19F-MRT wurden mit der PFC-Verbindung Perfluor-15-krone-5-ether durchgeführt, ein kleines ringförmiges Molekül, bei dem alle 20 Fluoratome chemisch äquivalent sind. Der Vorteil ist eine problemlose 19F-Bildgebung. Nachteilig ist jedoch die extrem lange biologische Halbwertszeit, so daß diese Substanz für repetitive Untersuchungen bzw. klinische Studien nicht in Frage kommt. Es sollen daher neue PFC-Verbindungen untersucht werden, von denen bekannt ist, daß sie nicht toxisch sind, über die Lunge abgeatmet werden und eine biologische Halbwertszeit von wenigen Tagen aufweisen (z.B. Perfluoroctylbromid, Perfluordecalin). Allerdings können diese PFCs aufgrund ihrer unterschiedlichen F-Kerne im Molekül nur mittels spektroskopischer Bildgebung artefaktfrei detektiert werden.
